The genus Rumex tuberosus L. subsp horizentalis belongs to one of the important family of flowering plant, polygonaceae, whose members are used as pharmaceutical herbs in traditional medicines in a large scale. Organogenesis and somatic embryogenesis were achieved by culturing of different explants (hypocotyl, cotyledon and epicotyl) excised from seedling of R. tuberosus. Embryogenic callus and adventitious buds(shoots) were induced on MS media supplemented by two levels sucrose (20 and 60 gr l1 and several combination of 2,4 Dichlorophenoxyacetic acid (2,4 D) + Kinetin (KIN) and (indole 3 acetic acid) IAA + 6 Benzylaminopurine (BAP) respectively. By augmentation of sucrose concentration from 20 gr l1 to 60 gr l1, the function of each mentioned hormonal mixtures, for embryogenic callus induction and shoot formation were reported more prosper. As it shown, maximum shoot generation were induced from hypocotyl explant in a medium containing 0.5 mg l1 IAA + 2.5 mg l1 BAP with 60 gr l1 sucrose and the medium supplemented by 5 mg l 1 2,4 D + 0.5 mg l 1 KIN with 60g l 1 sucrose has been reported for maximal root generation by application of hypocotyl as an explant. Further, extreme embryogenic calli were obtained from hypocotyl explant in a combination of 5 mg l 1 2,4 D + 0.5 mg l 1 KIN with 60 gr l 1 sucrose. Germinated and matured embryo were gained in MS medium containing 10 mg l 1 GA3 after 8 weeks of culture. In order to understanding embryo maturation process, changes in protein content during somatic embryogenesis was studied by spectrophotometric method. Result showed that, total protein content was increased during embryo development and it was gained to maximal level in advanced matured embryo. The synthesis and deposition of storage and Late embryogenesis abundant (LEA) protein during somatic and zygotic embryogenesis are usually regulated through ABA and water stress induce genes expression. Polyphenol oxidase and peroxidase activities were studied spectrophotometrically too. According to our research, whereas polyphenol oxidase activity was the most noticeable in preliminary embryo, the peroxidase activity, in matured advanced embryo was more remarkable than preliminary embryo and embryogenic callus. Either, total phenol and flavonoid content were determined by spectrophotometric method and they were reported in the extreme of ingredient, in natural plant. Also result proved, Total phenol content in regenerated plant and matured embryo were similar, approximately.
